Lion's mane is the one with a real route into the brain: its molecules stimulate nerve growth factor, the protein that keeps adult neurons alive 1. In older adults with creeping cognitive slippage, three grams a day lifted dementia-screening scores by week eight, and the gain faded a month after stopping 2. A later trial repeated the lift 3. It's a slow restorative, not a smart drug — and nobody has tested the healthy twenty-somethings who buy most of it.
Turkey tail has the strongest evidence and the narrowest use. Its PSK extract, added to chemotherapy for gastric cancer, cut the death hazard by about a tenth 4. That lives in oncology, under a clinician.
Cordyceps gives an untrained adult a small endurance bump after about three weeks of dosing 5. Reishi for sleep and chaga for longevity are the tradition-rich, trial-poor end: the popular sleep claim has essentially no human trial behind it 6.
The bottle is the whole decision, so read it before you read the dose.
If lion's mane works, it works slowly. Three or four months in, the foggy hour after lunch becomes an ordinary one, the name on the tip of your tongue arrives, mood settles a touch 2. Stop, and a month later the drift resumes. Nobody around you will comment on a transformed person. Cordyceps, if anything, shows up in the third mile of a run, not on race day 7. Reishi and chaga: expect to notice nothing.
Cheaper, better-studied options sit in every one of these lanes. For midlife cognition, creatine at three to five grams a day has a far larger trial base and costs less. For acute focus, caffeine with L-theanine. For the immune β-glucan itself, oats and barley carry the same fibre family in food. Mushrooms are a reasonable complementary play in one or two narrow cases, rarely the strongest move in their lane.
The fine print — when to skip it, and what people get wrong
"30% polysaccharides" can be 28% rice and 2% mushroom; assays find actual β-glucan from one to fifty percent of the labelled total 8. The five species are not interchangeable: reishi does not do lion's mane's job.
Skip on a blood thinner: reishi inhibits platelet stickiness 9. Skip on immunosuppressants; skip if pregnant. Long daily chaga tea has caused kidney injury 10; reishi has rare liver case reports 11.

Dual-extracted, β-glucan-assayed lion's mane or reishi at trial-equivalent doses runs ~$200–400/year; cheaper mycelium-on-grain products are mostly starch (McCleary & Draga 2016).
Lion's mane (Hericium erinaceus, 3 g/day) raised HDS-R cognitive scores over 16 weeks in adults with mild cognitive impairment (Mori et al. 2009) and improved cognitive function in older adults (Saitsu et al. 2019); the mechanism (erinacine-driven NGF synthesis, Kawagishi et al. 1994) is unique in the supplement space, but trials are small and Japanese-only.
Lion's mane reduced CES-D depression and indefinite-complaint scores in perimenopausal women over 4 weeks (Nagano et al. 2010); BDNF and NGF mechanism plausible (Spelman et al. 2017). Single small trial, replication thin.
One strong subdomain (PSK / turkey tail as oncology adjuvant; Oba et al. 2007, Eliza et al. 2012) anchors the upper end; the rest is small RCTs (lion's mane, cordyceps) or in-vitro and animal extrapolation (chaga, reishi sleep). Cochrane review of reishi cancer evidence rated low quality (Jin et al. 2016).
β-glucan-mediated immune markers (NK cell activity, CD3/CD4/CD8 counts) shift modestly with reishi and turkey-tail supplementation (Jin et al. 2016 Cochrane review; Torkelson et al. 2012), but consistent felt-experience improvements in healthy adults are not documented.
PSK (Trametes versicolor) reduces mortality hazard ~11% as a chemotherapy adjuvant in resected gastric cancer (Oba et al. 2007), but only in that clinical use under oncologist supervision. No mortality data for the rest of the category.
Cordyceps lifts ventilatory threshold ~10% in sedentary older adults (Chen et al. 2010) and improves time-to-exhaustion after 3 weeks of chronic dosing in untrained subjects (Hirsch et al. 2017); the effect is modest, slow, and concentrated in untrained populations.
Reishi's sleep claim rests on rodent NREM-architecture data and traditional convention (Wachtel-Galor et al. 2011); no human RCT establishes effects on sleep onset, duration, or quality. Score reflects the persistent traditional claim and weak mechanism, not trial-grade evidence.
